The McMaster technique uses a counting chamber which enables a known volume of fecal suspension (2 x 0.15 ml) to be examined microscopically. If the suspension is prepared using a precisely weighed aliquot of fecal material and a known volume of flotation solution, the number of eggs per gram (EPG) can be calculated.

Directions

Stirring vigorously (transfer solution to a flask to prevent spillage), take a sample with a pipette or medicine dropper and load one of the chamber of the slide. Repeat and fill other chamber. Wait 30 seconds for the eggs to float to the upper surface of the counting chamber.
